摘 要: | 目的建立测定人血浆中左卡尼汀(L-carnitine,LC)浓度的方法,并研究其药动学特性及生物利用度。方法采用双周期随机交叉试验设计[1]。运用高效液相色谱-质谱法(LCMS)检测血浆中药物血药浓度[2]。采用DAS Ver 2.0药动学软件对血药浓度时间数据进行处理,计算药代动力学参数。Tmax:Cmax、AUC 0-24 h及AUC 0-∞经对数转换后进行方差分析及双单侧t检验,进行生物利用度评价。结果参比制剂的主要药动学参数T1/2α为(2.06±1.20)h;T1/2β为(59.80±10.50)h;T max为(3.23±0.42)h,;AUC(0-∞)为(2518.3±310.65)μmol/(L.h);AUC(0-t)为(1311.54±218.32)μmol/(L.h);Cmax为(83.5±23.40)μmol/L。试验制剂的相对生物利用度为(98.1±11.6)%。结论该检测方法准确、灵敏、简便。符合血浆样品的测定要求,可以应用于血药浓度的测定和药代动力学研究。

Abstract: | Objective To establish the determination of human plasma concentration of L-carnitine ( L- earnitine, LC ), and study its pharmacokinetic characteristics and bioavailability. Methods A randomized crossover study of two-cycle design. High performance liquid chromatography - mass spectrometry (LCMS) detection in plasma drug plasma concentration[2]. DAS Ver 2.0 pharmacokinetics software processed to calculate the pharmacokinetic parameters of the plasma concentration-time data. Tmax:, Cmax, AUC 0 -24 h and AUC 0- ∞ by logarithmic transformation analysis of variance and two one-sided t-test, evaluation of bioavailability. Results Reference preparation pharmacokinetic parameters T1/2α (2.06± 1.20) h; T1 / 2 β (59.80 ± 10.50) ; T max (3.23 +0.42)h; AUC (0 -∞ ) (2518.3 ±310.65) μmol/L-1 oh; AUC (O-t) (1311.54 ±218.32) μmol/L-1 o h; C max (83.5 ±23.40) μmol/L-1. The relative bioavailability of the test preparation (98. 1± 11.6)%. Conclusions The detection method is accurate, sensitive and easy. Determination of plasma samples requirementscan be applied to the determination of plasma concentration and pharmacokinetic studies. |
