生何首乌、制何首乌对大鼠肝微粒体CYP450的影响

摘    要:目的研究肝微粒体细胞色素P450(CYP450)以及血液生化酶在生何首乌、制何首鸟致SD大鼠肝损伤中的作用。方法将SD大鼠分为3组,每日灌胃给药30g/kg,连续给药90d,末次给药后处死,称取脏器指数,测定肝脏CYP450、血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、白蛋白(ALB)、肌酐(CREA)、尿素氮(BUN)、甘油三酯(TG)等含量。结果生何首乌组肝微粒体蛋白显著性下降,CYP450含量显著增高(P〈0.01),制何首乌组无明显变化;血液生化酶中生何首乌组的血清ALT、AST显著性降低,制何首乌血清ALP显著性升高;生何首乌和制何首乌的血清TP、TG均显著性降低;血清BUN中二者均呈下降趋势,但生何首乌具有显著性;CREA二者均呈显著性下降趋势。结论生何首乌致肝损伤并有CYP450的明显升高。制何首鸟对CYP450无影响。生何首乌和制何首乌能够影响血液生化酶,生何首乌减低ALT、AST,制何首乌升高ALP,二者同时降低TG、BUN和CREA,这意味生着何首乌、制何首乌致大鼠肝物质代谢途径的作用机制有所不同。

Effect of Unripe Radix Polygoni Multiflori,Radix Polygoni Multiflori on Rat Liver Microsomal CYP450

Abstract:Objective to study the liver microsomal cytochrome P450 (CYP450) and blood biochemical enzyme in unripe Radix Polygoni Muhiflori, Radix Polygoni Multiflori on liver injury induced by SD in rats. Methods SD rats were divided into 3 groups, daily intragastric administration of 30 g/kg, continuous administration of 90d, put to death after the last drug, weighed the organ index, determinate the liver CYP450, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin (ALB), creatinine ( CREA), blood urea nitrogen (BUN), triglyceride (TG) con- tent. Results unripe Radix Polygoni Muhiflori group of liver microsomal protein was significantly decreased, CYP450 con- tent increased significantly ( P 〈0.01 ), Radix Polygoni Multiflori group had no obvious change; the crude enzymes in the blood serum ALT, AST were significantly lower in unripe Radix Polygnni Multiflori, serum ALP increased significant- ly; TP, TG were significantly decreased; two serum BUN showed a decreasing trend, but the crude was significant; CREA were all decreased significantly. Conclusion unripe Radix Polygonum Multiflori induced liver injury and CYP450 was significant raised. Radix Polygoni Multiflori had no effect on CYP450. Unripe and Radix Polygoni Muhiflori can affect the blood biochemical enzyme, the crude ALT, AST decreased, Radix Polygnni Multiflori ALP increased , both of them reduced the TG, BUN and CREA, the mechanism that the unripe Radix Polygoni Muhiflori, Radix Polygoni Muhiflori in- duced liver metabolism pathway is different.
