摘    要:目的探讨磷酸肌酸钠(CP)对急性心肌梗死(AMI)后大鼠心律失常及心室肌细胞钠电流(INa)的影响。方法采用大鼠心肌缺血再灌注致大鼠心律失常动物模型,以BL-420生物机能实验系统观察ECG的相关指标,并进行统计分析。大鼠开胸左前降支结扎造AMI,酶解分离心室肌细胞,采用全细胞膜片钳记录技术记录左前降支供血区心外膜细胞的INa。结果①CP组与对照组相比较,心肌缺血再灌注大鼠模型的室性早搏和室速均明显减少(n:10,P〈0.05)。②应用cP(20mg/kg)后与AMI组比较,INa峰值从(-4.82±1.91)nA下降到(-2.56±1.59)nA,差异有统计学意义(P〈0.01,n=6)。结论①CP对大鼠心肌缺血再灌注心律失常具有保护作用。②CP可显著降低AMI大鼠心室肌细胞钠电流的幅值。

Abstract:Objective To study the effect of Creatine phosphate (CP) on the arrhythmia and the sodiumcurrent of rat isolated ventricular myocytes after the acute myocardial infarction. Methods Rat arrhythmia model was induced by myocardial ischemia reperfusion, the rats were divided into control group and CP group, BL-420 biologica/ functional experimental system was used to observe the relevant indicators of ECG. Whole cell patch clamp technique was applied to record the sodium current of rat isolated ventricular myoeytes from ischemie zone adjacent to infarcted area. Results (1)The ventricular premature beat and ventricular tachycardia that induced by myocardial ischemia reperfusion decreased significantly in CP group compared with control group. (2) I Na of CP (20 mg/kg)deereased from(-4.82±l.91 )nA to (-2.56±1.59)nA, there was significant difference compared to the acute myocardial infarction group (P〈0.01 ,n=6). Conclusion CP has a remarkable protecting effect on experi- mental arrhythmia and CP inhibit the sodium current of rat isolated ventrieular myoeytes after the acute myocardial infarction remarkably.
