血管紧张素-(1-7)对野百合碱诱导的肺动脉高压大鼠炎症反应的作用

摘    要:目的 探讨血管紧张素-(1-7) Ang-(1-7)]对野百合碱(MCT)诱导的肺动脉高压大鼠炎症反应的作用.方法 雄性SD大鼠30只,随机分为对照组、MCT组和Ang-(1-7)组.MCT组和Ang-(1-7)组一次性颈部注射MCT 60 mg/kg,24 h后经微泵持续泵入生理盐水或Ang-(1-7) (24 ?滋g·kg-1·h-1).对照组一次性颈部注射相同体积的生理盐水,24 h后泵入生理盐水.4周后,测定大鼠的右室收缩压(RVSP)和右心肥厚指数(RVHI),测定肺小动脉管壁厚度(WT)占动脉外径(ED) 的百分比(WT%)及管壁面积(WA)占血管总面积的百分比(WA%).采用免疫酶联反应(ELISA)检测白细胞介素-6 (IL-6)、肿瘤坏死因子-?琢(TNF-?琢)及单核细胞趋化蛋白-1(MCP-l)的浓度.结果 野百合碱诱导4周后,MCT组与对照组相比,RVSP、RVHI、WT%、WA%明显升高,肺动脉压力明显升高,肺组织出现了明显的重构,同时IL-6、TNF-α和MCP-l的含量也明显升高.Ang-(1-7)组与MCT组相比上述指标明显降低.结论 在野百合碱诱导的肺动脉高压模型中,Ang-(1-7)可能通过降低IL-6、TNF-α和MCP-l的含量,抑制炎症反应,预防肺动脉高压的发生.

Effects of Ang-(1-7) on inflammation in pulmonary arterial hypertension rat induced by monocrotaline

Abstract:Objective To investigate the effects of Ang-(1-7) on inflammation in pulmonary arterial hypertension rat induced by monocrotaline. Methods Thirty SD rats were randomly divided into three groups: control group, MCT group and Ang-(1-7) group. Rats in MCT group and Ang-(1-7) group received 60 mg/kg MCT injection subcutaneously and after 24 hours received either saline or 24 kg·kg-1·h-1 of Ang-(1-7) injection via osmotic minipumps for four weeks. These rats in control group were firstly injected saline subcutaneously and then received saline injection via osmotic minipumps for four weeks. Right ventricular systolic pressure (RVSP) and right ventricular hypertrophy index (RVHI) were measured. Percentage of wall thickness (WT%) and percentage of wall area (WA%) of pulmonary arterioles were evaluated. Concentration of interleukin-6(IL-6), tumor necrosis factor-α (TNF-α) and monocyte chemotactic protein-1 (MCP-1) were measured by ELISA. Results Four weeks after monocrotaline injection, compared with control group, RVSP, RVHI, WT%, WA% and concentration of IL- 6, TNF-α and MCP-lwere significantly increased in MCT group. Compared with MCT group, the above parameter were significantly decreased in Ang-(1-7) group. Conclusion Ang-(1-7) might prevent the development of pulmonary arterial hypertension induced by monocrotaline. The reason might be that Ang-(1-7) might decrease inflammatory response and decrease the concentration of IL-6, TNF-α and MCP-1.
