阿托伐他汀对自发性高血压大鼠心肌组织GLUT4表达的影响及意义

摘    要:目的:探讨阿托伐他汀对自发性高血压大鼠(spontaneously hypertensive rats,SHR)心肌组织葡萄糖转运蛋白4(glu-cose transporter-4,GLUT4)表达的影响及其对心肌肥厚的逆转作用与可能机制。方法:将16只8周龄SHR随机分为:阿托伐他汀药物干预组(ATV组,n=8)与SHR模型对照组(SHR组,n=8),以8只同周龄Wistar-Kyoto大鼠作为正常对照(WKY组)。观察阿托伐他汀灌胃10周后大鼠血脂、心肌肥厚指标、GLUT4 mRNA及其蛋白表达的改变。结果:经过10周治疗,ATV组及SHR组血脂水平差异无统计学意义(P>0.05)。ATV组左室重量指数低于SHR组(P<0.05)。在ATV组,GLUT4表达高于SHR组(P<0.05)。结论:阿托伐他汀可上调SHR肥厚心肌组织中GLUT4的表达,有效逆转左室肥厚。

Abstract:Objective:To investigate the effects of atorvastatin on GLUT4 expression in myocardial tissue of spontaneously hypertensive rats(SHR) and its reverse effect and possible mechanism of cardiac hypertrophy.Methods: 16 eight-week-old male spontaneously hypertensive rats were randomly divided into two groups: Group of atorvastatin intervention(ATV,n= 8),and group of SHR model comparison(SHR,n=8).8 Wistar-kyoto rats with the same age were served as the normal control(WKY,n=8).The groups were intragastric administration by atorvastatin for 10 weeks,and the change of blood lipid,the index of cardiac hypertrophy and GLUT4 mRNA protein expression will be observed.Results: There was not obvious difference of blood lipid level between ATV and SHR group(P>0.05) after 10 week’s treatment.Compared with SHR group,the index of left ventricular mass is lower in ATV group(P<0.05).The GLUT4 expression of ATV group is higher than SHR group(P<0.05).Conclusion: Atorvastatin increases GLUT4 expression in SHR cardiac hypertrophy and effectively reverses left ventricular hypertrophy of spontaneously hypertensive rats.
