二硫化碳对大小鼠睾丸组织超微结构影响的研究

摘    要:
目的 从超微结构损伤的角度观察、探讨二硫化碳(CS2)不同浓度染毒对雄性大、小鼠睾丸组织各级生精细胞及支持细胞、间质细胞的影响.方法 将40只雄性Wistar大鼠和32只雄性昆明小鼠分别随机分为对照组和不同CS2染毒浓度3组(大鼠:50、250、1 250 mg/m3,小鼠:220、660、1 980 mg/m3),静式吸入染毒,2 h/d,5 d/周,共10周.染毒结束后,取大、小鼠睾丸组织,透射电镜观察大、小鼠睾丸组织超微结构的改变.结果大鼠50 mg/m3和250 mg/m3及小鼠220 mg/m3和660 mg/m3浓度组精原细胞、精母细胞胞质中可出现空泡,线粒体轻度肿胀.大鼠1 250 mg/m3和小鼠1 980 mg/m3浓度组,间质细胞及支持细胞胞质中出现明显空泡;精原细胞、初级精母细胞胞质中空泡较多,线粒体肿胀加重;精子细胞胞质中也出现空泡,但线粒体肿胀较轻;精子"9*2+2" 轴丝结构紊乱,部分微管和致密纤维溶解缺失,有的甚至完全断裂.结论吸入一定浓度的CS2可使大、小鼠睾丸组织各类生精有关细胞的内质网、各级生精细胞的线粒体及精子尾部轴丝等超微结构发生病理变化.

Abstract:
Objective To observe and approach the effects of CSz on the ultrastructure of all the spermatogen- ic cells,sertoli ceils and Leydig cells in the testis of rats and mice under different concentrations. Methods 40 male Wistar rats and 32 mate Kunming mice were randomly divided into a control group and three CSz concentration groups (rats.. 50,250,1 250 mg/m3 ; mice: 220,660,1 980 mg/m3 ) respectively. Six experimental groups were exposed to CS2 by inhalation for 10 weeks with 2 h/d, 5 d/w. After 10-week treatment, take their testes, use transmission electron microscope ( TEM ) to observing the ultrastructure of the testis cells. Results In rats 50 mg/m3 ,250 mg/ m~ and mice 220 mg/m3 ,250 mg/m3 concentration groups, the mitochondria in spermatogoniums and spermatocytes were swelled slightly. In rats 1 250 mg/ma and mice 1 980 mg/m3 concentration groups, there are obvious vacuolus in the endochylema of Leydig cells and sertoli cells; the vacuolus in the endochylema of spermatogoniums and spermat- ocytes were increased, the mitochondria in them were exacerbated swelled; vacuolus appear to occur in the en- dochylema of spermatids,but the swelling degree of the mitochondria were lighter; the"9 * 2+.2" structure of the flagellae of spermatozoa were disordered, some microtubules and feltwork were dissolved or missed, some have even broken completely. Conclusion The ultrastructure of the endoplasmic reticulum in all the kinds of testis spermato- genesis related cells, the mitochondria in spermatogenic cells, and the axial filament of the flagellae of spermatozoa, may change pathologically when the rats or mice inhaling a certain concentration of CSz.
