苦参素与顺铂联合应用诱导肝癌SMMC-7721细胞凋亡及其机制

摘    要:目的:探讨苦参素和顺铂联合应用诱导人肝癌SMMC-7721细胞凋亡及其机制。方法:采用MTT法检测细胞增殖抑制率,流式细胞仪检测细胞凋亡率,应用金氏公式分析药物间的相互作用,RT-PCR方法检测fas和caspase-3基因mRNA表达水平。结果:与单药组相比,联合用药对肝癌SMMC-7721细胞增殖抑制率明显增高(P<0.05),细胞凋亡率显著增高(P<0.05),并表现明显的协同作用,细胞fas和caspase-3基因mRNA表达水平明显上升(P<0.05)。结论:苦参碱与顺铂联合应用能明显诱导肝癌SMMC-772 l细胞凋亡并有协同作用,其机制可能与上调细胞fas和caspase-3基因表达有关。

Abstract:Objective: To investigate the apoptosis-inducing and its mechafiism of matrine combined with DDP on human hepatocarcinoma SMMC-7721 cells.Methods:The proliferation inhibitory rate was evaluated bv MTT assay.Induction of cell apoptotic rate was determined by flow cytometry.The interaction between Matrine and DDP was analyzed by means of Jin′s formulation.The expression of fas and caspase-3 mRNA was detected by semi-quantitive RT—PCR.Results:Compared with single drug group,the inhibitory rate of combined group was increased obviously(P<0.05),the apoptotic rate was increased markedly(P<0.05),and their synergistic effect could be observed.The expression of fas and caspase-3 mRNA was increased significantly(P<0.05).Conclusions:Matrine combined with DDP could obviously induce apoptosis of human hepatocarcinoma SMMC-7721 cells and display the synergistic effect,which may be involved in increasing the expressions of fas and caspase-3 gene.
