| 摘 要： | 应用逆转录多聚酶链式反应（ＲＴ－ＰＣＲ）技术，自行设计并合成一对引物，成功地从活化的正常人外周血单个核细胞中扩增出ｈＩＬ－１０ｃＤＮＡ基因。并定向插入ｐＵＣ１８载体中，经ＤＮＡ序列测定最终确定为ｈｌＬ－１０ｃＤＮＡ基困，这将为令后ｈＩＬ－１０基因探针的制备和基因表达，进一步在基因水平上探讨ｈＩＬ－１０的生物学功能提供了物质基础。 |

| Abstract: | With reference to the human IL-10 gene in GenBank database, a pair of DNA primers (27 bases and 20 bases) was designed and synthesized, A 610 bp fragment containing hIL-10gene was successfully amplified by using RT-PCR and then cloned into pUC18 plasmid, constructing a recombinant plasmid-pUhIL-10. The recombinant plasmid pUhIL-10 was selected and sequenced. The result showed that the fragment cloned in pUhIL-10 was hIL-10 cDNA, it would promote the further research in molecular biology of hIL-10. |
