熟地强筋合剂对大鼠骨髓基质细胞向成骨细胞分化的影响

摘    要:目的:观察熟地强筋合剂在骨髓基质细胞向成骨分化过程中的作用。方法:采用体外培养大鼠骨髓基质细胞,分别观察各含药血清组碱性磷酸酶染色、Ⅰ型胶原染色情况;取各组不同时间点培养液测定碱性磷酸酶比活性及骨钙素含量。结果:碱性磷酸酶染色强弱顺序为D组>C组>B组,Ⅰ型胶原染色强弱顺序为D组>C组>B组>A组,碱性磷酸酶比活性各加药组与对照组比较明显提高(P<0.05);各加药组与对照组比较BGP含量均明显提高。结论:熟地强筋合剂能促进大鼠骨髓基质细胞增殖,诱导大鼠骨髓基质细胞向成骨细胞分化。

The Effect on Differentiation to Osteoblast of SD Rat'S Bone Marrow Stromal Cells in vitro by Shu Di Qiang Jin Mixture

Abstract:Objective:To observe the differentiation to osteoblast of SD rat’ s marrow stromal cell cultured by Shu Di Qiang Jin Mixture.Methods:The serum pharmacology method was used to culture the SD rat’ s marrow stromal cell.The alizarin Bordeaux starting,Van-GiegonⅠcollagen staining method were used to observe the expressions of ALP and typeⅠcollagen respectively on different collection time.Results:The intensity of ALP dyeing sequenced as:D>C>B,and collagenⅠdyeing sequenced as:D>C>B>A.The alkaline phosphatase activity and the amount of bone glaprotein were higher in B,C,D groups compared with the control group at the same time(P<0,05).Conclusion:Shu Di Qiang Jin Mixture could accelerate the proliferation of the BMSCs,and could induce the BMSCs differentiation,accelerate the proliferation and differentiation of the BMSCs.
