摘    要:目的:探讨慢性低O2高CO2对小鼠皮层及海马神经细胞凋亡及硝基酪氨酸表达的影响.方法:雄性C57BL/6小鼠24只,随机分为对照组、低O2高CO22 w组和低O2高CO24 w组,采用原位末端标记法检测皮层和海马区神经细胞凋亡并计算凋亡指数(AI),免疫组织化学染色检测皮层和海马区细胞硝基酪氨酸的表达.结果:慢性低O2高CO22 w组和4 w组小鼠皮层和海马AI及硝基酪氨酸表达均明显高于对照组,两者呈正相关.结论:慢性低O2高CO2可以诱发小鼠皮层和海马神经元凋亡,可能与其促进硝基酪氨酸表达有关.

Neurons apoptosis and nitrotyrosine expression in cortex and hippocampus induced by chronic hypoxic hypercapnia

Abstract:Objective: To explore the effects of chronic hypoxic hypercapnia on the apoptosis of mouse neural cells and the expression of nitrotyrosine in the cortex and hippocampus. Methods: Twenty four male C57BL/6 mice were randomly divided into three groups: normal control, hypoxic hypercapnia 2 w, hypoxic hypercapnia 4 w. The apoptosis indexes (AI) of neural cells in cortex / hippocampus were achieved by terminal deoxynuleotidyl transferase-mediated dUTP nick end labeling (TUNEL)and the expression of nitrotyrosine in the cortex / hippocampus was detected by immunohistochemistry. Results: Both the AI and the expression of nitrotyrosine of neural cells in cortex and hippocampus in hypoxia hypercapnia groups were higher than those in normal control group. Correlative analysis showed that there was a positive correlation between AI and the expresson of nitrotyrosine. Conclusion: Chronic hypoxic hypercapnia can induce neural cells apoptosis in mouse cortex and hippocampus,which may be associated with its effects on expression of nitrotyrosine.
