白藜芦醇对于外周血内皮祖细胞体外增殖分化的影响

摘    要:
目的:观察白藜芦醇对内皮祖细胞体外扩增分化过程中内皮系基因的表达及粘附、迁移功能的影响。方法:密度梯度离心法获得人外周血单个核细胞,在向内皮祖细胞诱导分化的同时,加入不同浓度的白藜芦醇(1、5、15、60μM)干预7天,随后采用流式细胞术观察CD34、CD31、KDR基因的表达;用RT-PCR和ELISA方法检测干预后内皮型一氧化氮合成酶的表达;同时测定干预后细胞的粘附和迁移功能。结果:低浓度白藜芦醇(5μM)不同程度促进内皮祖细胞增殖分化中CD34、CD31、KDR的表达,上调eNOS的mRNA和蛋白水平表达,并促进内皮祖细胞粘附和迁移功能;而高浓度白藜芦醇(60μM)则不同程度下调CD34、CD31、KDR、eNOS的表达和抑制粘附、迁移功能。结论:低浓度白藜芦醇可促进外周血内皮祖细胞内皮系基因的表达,并且促进其粘附和迁移,而高浓度则产生相反的作用。

Abstract:
Objective: To investigate the effect of resveratrol on endothelial cell-specific gene expression of endothelial progenitor cells (EPCs) during proliferation and differentiation in vitro as well as its adhesion and migration function.Method: Peripheral blood monocytes (PBMCs) were isolated by ficoll density gradient centrifugation from peripheral blood and cultured in VEGF, bFGF supplemented medium, simultaneously with the addition of resveratrol (1,5,15,60μM) for 7days. Then expressions of CD34, CD31, KDR were determined by flow cytometry analysis. eNOS expression was assayed by RT-PCR and ELISA. We also analysed the adhesion and migration function of EPCs after resveratrol interference.Results: Low concentration (5μM) of resveratrol could increase the expression of CD34, CD31 and KDR in attached cells and upgrade the eNOS expression; while high concentration (60μM) decreased the expression of aforementioned markers. In addition, low concentration promoted adhesion and migration function of EPCs, while high concentration attenuated above functions.Conclusion: Low concentration of resveratrol facilitates EPCs proliferation and differentiation toward endothelial lineage as well as promotes its adhesion and migration function, but high concentration exerts adverse effects.
