滋肾通络方对高糖诱导的大鼠肾小球系膜细胞TGF-β1、MMP-2、MMP-9mRNA表达影响的实验研究

摘    要:目的:观察滋肾通络方含药血清对高糖环境下LPS刺激大鼠肾小球系膜细胞(GMCs)TGF-β1、MMP-2、MMP-9mRNA的影响,并探讨其在防治糖尿病肾病(DN)中的机制。方法:以高糖环境下LPS刺激大鼠肾小球系膜细胞生长建立模型,分为以下8组:(1)正常组;(2)高糖组;(3)高糖+LPS组;(4)苯那普利组;(5)滋肾通络方小剂量组;(6)滋肾通络方中剂量组;(7)滋肾通络方大剂量组。采用RT-PCR技术从分子生物学水平检测各组系膜细胞中TGF-β1、MMP-2及MMP-9mRNA表达。结果:与正常组相比,高糖组、高糖+LPS组TGF-β1mRNA的表达明显升高(P<0.01),MMP-2及MMP-9mRNA的表达明显降低(P<0.01);与模型组相比较,滋肾通络方大、中、小剂量组对大鼠肾小球GMCs TGF-β1mRNA表达显著降低(P<0.05),MMP-2、MMP-9mRNA表达明显升高(P<0.05)。结论:滋肾通络方含药血清能够抑制大鼠肾小球GMCs TGF-β1mRNA的表达,增加MMP-2、MMP-9mRNA的表达,从而通过增加肾小球系膜细胞细胞外基质的降解以延缓DN进展。

Abstract:To observe the influence of the capsule of zishentongluo decoction(ZSTL) to TGF - β1 ,MMP -2, MMP-9mRNA of glomerular mesangial cells (GMCs) induced by LPS in high glucose condition, and explore its possible mecha- nism. Methods: Build the growth model of podocyte induced by LPS in high glucose condition, divide into 8 groups : ( 1 ) Normal group ; ( 2 ) High glucose group; ( 3 ) High glucose plus LPS group; (4) Benazepril group; ( 5 ) Low - dose group of Zishentongluo Decoc- tion(ZSTL) ; (6) Middle - dose group of Zishentongluo Decoction (ZSTL) ; (7) Large - dose group of Zishentongluo Decoction ( ZSTL). Detect glomerular mesangial cells TGF - 13t, MMP - 2,MMP - 9mRNA expression of each group by RT - PCR on molecular biology level. Results:Compared with the normal group, the expression of glomerular mesangial cells (GMCs) TGF - {31 mRNA in- creased definitely (P 〈0.01 ), MMP- 2,MMP- 9mRNA decreased definitely (P 〈 0.01 ) in high glucose group and high glucose plus LPS group. Compared with the high glucose group and high glucose plus LPS group, the expression of glomerular mesangial cells (GMCs) TGF - β1 mRNA decreased ( P 〈 0.05 ), MMP - 2,MMP - 9mRNA decreased definitely ( P 〈 0.05 ). Conclusion : Zishent- ongluo decoction(ZSTL) can reduce glomerular mesangial cells TGF- [31 mRNA expression and increase MMP- 2,MMP -9mRNA expression, by increasing extracellular matrix degradation in order to delay the the DN progress.
