大鼠正畸牙移动张力侧牙周组织中TGF-β_1蛋白表达的研究

摘    要:
目的检测正畸牙移动张力侧TGF-β1蛋白表达变化及时间分布特点,初步探讨正畸牙移动中牙槽骨改建与TGF-β1的关系。方法45只雄性S.D.大鼠随机分为正常对照组,牙移动12h组、1d组、2d组、3d组、5d组、7d组、14d组、21d组,通过对大鼠上颌第一磨牙的近中移动,运用免疫组织化学方法,检测了正畸牙移动张力侧TGF-β1蛋白表达强度及时间分布特点。结果牙移动1d后,张力侧牙周膜区TGF-β1免疫组化染色阳性明显增强,2d后达到高峰,牙移动2d后,牙槽骨表面TGF-β1蛋白染色强度明显增强,5d后达到高峰。结论正畸牙移动过程中,在机械力作用下TGF-β1作用参与了张力侧的骨形成。

Abstract:
Objective To examine the protein localization of transforming growth factor-β1 (TGF-β1) in tension sides during orthodontic tooth movement. Methods 45 adult male Sprague-Dawley rats were randomly grouped into 9: the control and the 8 groups for experimental tooth movement. The rats were treated with the fixed orthodontic appliance by 50 gram forces that moved medially the maxillary first molars. Then the animals were sacrificed at the corresponding treatment times. The TGF-β1 expression was examined by immunohistochemically staining. Results The expression of the mRNA and of TGF-β1 was weak in control rats. After 1 day of tooth movement, intense immunostaining for the protein of TGF-beta1 was observed in the periodotium and reach the maximum in 2 d; after 2 days of tooth movement, intense immunostaining for TGF-β1 was observed in the cubically shaped osteoblasts and reach the maximum in 5 days. Conclusion TGF-β1 as a local regulator plays an important role in periodontium remodeling during tooth movement.
