血清直接、间接胆红素对氧化酶法测定甘油三脂浓度的影响分析

摘    要:
目的通过分析不同种类胆红素对甘油三脂(triglyceride,TG)氧化酶测定方法的干扰,探讨直接胆红素(direct bilirubin,DBIL)、间接胆红素(indirect bilirubin IBIL)对过氧化氢酶(H2O2)偶联Trinder反应的TG氧化酶法测定结果影响程度的不同。方法我们制备系列不同浓度的DBIL、IBIL混合血清,且根据TG含量分为高、中、低三组,其中DBIL、IBIL均在日立7170A上检测;而每种血清标本中TG含量均经氧化酶(GPO-PAP)和甘油三脂乙酰丙酮显色法检测。结果DBIL对TG检测的干扰大,当其浓度达280μmol/L时高、中、低三组标本中TG回收率仅为62.8%、60.3%、58.8%;而IBIL对TG检测的干扰小,当浓度达240μmol/L时TG回收率仍达90%以上。结论DBIL、IBIL对TG氧化酶测定结果的影响程度不同在实际操作中应区别对待。尤其在DBIL含量较高的标本用氧化酶法测TG时产生的负干扰,在结果审核时应重视加以校正。

Abstract:
Objective To examine the effect of different kinds of bilirubin that include direct bilirubin and indirect bilirubin on the direct enzymatic measurement of triglyceride(TG)by using the TG and peroxidase couple reaction.Methods we prepared much mixed sera of the different level of direct bilirubin(DBIL)and indirect bilirubin(IBIL),and they were divided three groups of high、middle and low according to the level of TG.The level of DBIL and IBIL were measured by HITACHl 7170A automatic analyzer,the level of TG of each serum sample was measured by using both the direct enzymatic measurement and acetic acetone measurement.Results DBIL produced greatest negative interference,when concentration of DBIL was 280 μmol/L,recovery rate of TG was only 62.8%、60.3%、58.8%,but the effect of IBIL was slight,when concentration was 240 μmol/L,recovery rate of TG was over 90%.Conclusion The DBIL and IBIL produced different interference in the enzymatic measurement of TG,They should be distinguished in our every day work.Especially,the interference produced by the higher lever of DBILwas greater,was adjusted in the result of checking.
