姜黄素联合MMC对宫颈癌HeLa细胞生长抑制作用观察

摘    要:目的:比较姜黄素分别和中、低剂量丝裂霉素C(MMC)联用与高剂量MMC单用对宫颈癌He—La细胞的抑制作用,探讨两药联用的效果是否等同甚至优于高剂量MMC单用的效果,为临床上开展较低剂量MMC与姜黄素联用治疗宫颈癌,以减少MMC用药剂量、降低其毒副作用提供实验依据。方法:将姜黄素与不同剂量的MMC联用或单独作用于体外培养的HeLa细胞,以MTT法检测给药后各组HeLa细胞的生长抑制情况。结果:MTT结果显示,姜黄素联合MMC对人宫颈癌HeLa细胞的生长具有明显的抑制作用,并呈剂量一时间依赖关系。姜黄素联合低剂量MMC组与高剂量MMC单用组对HeLa细胞的抑制作用无显著差异(P〉0.05),而姜黄素联合中剂量MMC组对HeLa细胞的抑制作用显著高于高剂量MMC单用组(P〈0.05)。结论:姜黄素与中剂量MMC联用能有效抑制宫颈癌HeLa细胞的生长,为临床上宫颈癌的化疗提供了一个新方法。

Abstract:Objective: To compare the Growth Inhibitory effect of low-dose mitomycin C (MMC)combined with curcumin on the cervical carcinoma cell line HeLa with high-dose MMC alone, and to understand the synergistic effect of two drugs is equal to or bet- ter than the high-dose of MMC single effect. Methods: The HeLa cells cultured in vitro were treated with various dose of MMC combined with curcumin and high-dose MMC alone for different periods (24h,48h), the inhibitory effects of MMC and curcumin on growth of HeLa cells were measured by the MTT assay. Results : MTT test show that the growth of HeLa cells was suppressed by MMC and curcumin significantly in dose and time dependent manner, and there is no significant difference in grouth inhibition rate of HeLa cells between the low-dose MMC combined with curcumin group and the high-dose MMC group(P〉0.05). In addition, grouth inhibition rate of HeLa cells in curcumin combined with medium dose MMC group was significantly higher than that of high-dose MMC alone group (P〈 0. 05). Conclusion: This study suggests that low dose MMC combined with curcumin could inhibite the grouth of HeI.a cells effectively, providing a new option for the cervical carcinoma chemotherapy.
