| 摘 要： | 目的 为了筛选抗乙型肝炎病毒(HBV)药物的细胞模型。本研究探索了转基因细胞2．2．15的培养条件及分泌表达的动力学变化。方法 用ELISA法动态检测2．2．15细胞分泌表达HBsAg和HBeAg的动力学变化。用PCR技术检测细胞和细胞培养上清中HBV-DNA。结果 2．2．15细胞在适宜的培养条件下能够持续分泌表达HBsAg和HBeAg，培养第6天达分泌高峰。第7天有所下降。PCR结果证明细胞和培养液上清中HBV-DNA为阳性。结论 转基因细胞2．2．15可作为筛选抗HBV药物的细胞模型。

| Abstract: | Objective To screen the cell model of anti-HBV drugs, we studied the cultural condition and kinetic
changing of the secretary expression of 2. 2. 15. Methods We detected kinetic changing on the secretary expressino of HBsAg and HBeAg in the 2. 2. 15 medium supernatant. PCR was used to detect HBV-DNA in 2. 2. 15 cell and 2. 2. 15 medium surpernatant. Results The cell of 2. 2. 15 was able to consecutively secrete HBsAg and HBeAg in the best cultural condition. HBsAg and HBeAg reached the highest peak at the 6th day and began to decline at the 7th day after culturing . HBV-DNA in 2. 2. 15 cell and 2. 2. 15 medium surpernatant were positive by PCR. Conclusion 2. 2. 15 cell would be an ideal cell model to screen the anti-HBV drugs. |
