摘    要:
目的:探讨阿蒂莫耶番荔枝内酯对有多药抗药性的阿霉素的人乳腺癌细胞MCF-7/ADR凋亡的诱导作用,揭示其抗癌机制。方法;用MTT法观察药物对细胞的生长抑制作用,TUNEL法,流式细胞仪及光镜,电镜等技术研究癌细胞凋亡。结果;药物对癌细胞有明显生长抑制作用,IC50O 10.5mg/L;光1镜,电镜可见染色质浓缩,聚集核膜下,核碎裂等改变;流式细胞仪检测到凋亡峰;10mg/L药物作用48小时,TUN

Abstract:
Objective: To investigate the induction of apoptosis of human breast cancer cell line MCF-7/ADR by Annonaceons acetogenins (AA) in order to demonstrate the mechanism of its anticancer action. Methods: MTT method, TUNEL, flow cytometry (FCM), light microscopy and transmission electronmicroscope were used. Results: MCF-7/ADR treated with AA underwent cell proliferation inhibition, IC50 is 10.5 mg/L; cell shrinkage, condensation cell chromatin clinged to periphery of the nuclear and fragmentated can be detected by light microscopy and transmission electronmicroscope, apopttotic peaks were also detected on FCM, the TUNEL-positive cells were 57.3% after being treated with AA at 10 mg/L for 48 hours. Conclusion: AA can induce apoptosis of MCF-7/ADR cell line.
