Abstract:
Objective To study the effect of bone morphogenetic protein 4 (BMP4) on the proliferation, migrationand invasion of breast cancer cell line MCF-7 in vitro. Methods MCF-7 cells were infected with lentivirus Lv-BMP4and lentivirus Lv-shBMP4 to construct MCF-7/Lv-BMP4 overexpression group and MCF-7/Lv-shBMP4 interferencegroup. There were 4 groups in this experiment: Blank group, Lv-NC group, Lv-BMP4 group and Lv-shBMP4 group.RT-PCR and ELISA were used to detect BMP4 overexpression and interference in MCF-7 cells. CCk-8 assay wasused to detect the proliferation of MCF-7 cells. Scratch healing and Transwell were used to detect the migration andinvasion ability of MCF-7 cells. Results RT-PCR and ELISA showed that the BMP4 over-expressed breast cancer celllines MCF-7/Lv-BMP4 and the endogenous BMP4 down-regulated cell lines MCF-7/Lv-shBMP4 were successfullyconstructed. The CCK-8 results showed that the cell proliferation in BMP4 over-expressed group was significantlyinhibited but the cell proliferation significantly low in MCF-7/Lv-shBMP4 group when compared with the controlgroup. Wound-healing assay found that the scratch healing rate in Lv-BMP4 group was significantly lower than thatin the control group after 48 h,while the scratch healing rate in Lv-shBMP4 group was significantly higher than thatin the control group. Transwell migration assay showed that MCF-7 cells in Lv-BMP4 group getting through a fewfilms after 48 h was significantly lower than that in the control group, while the migrated number of MCF-7 cells inLv-shBMP4 group was significantly higher. Conclusion BMP4 can inhibit the proliferation and migration of breastcancer MCF-7 cells in vitro.
