羟基红花黄色素A对高糖作用下恒河猴脉络膜-视网膜血管内皮细胞迁移及VEGF表达的影响

摘    要:目的 研究羟基红花黄色素A(HSYA)对高糖作用下恒河猴脉络膜-视网膜血管内皮细胞(RF/6A)迁移的影响,探讨HSYA抑制视网膜脉络膜新生血管内皮细胞的作用机制。方法 采用细胞划痕修复实验和半定量RT-PCR方法,检测HSYA对正常和高糖作用下RF/6A迁移和VEGF表达的影响。结果 HSYA在18 mg/L,37 mg/L,73mg/L浓度对高糖(22mmol/L)诱导下RF/6A细胞有显著抑制作用,而对正常糖(5mmol/L)下RF/6A细胞无显著抑制作用;与正常糖组比较高糖组VEGF明显表达,而加HSYA组VEGF表达降低。结论 一定浓度的HSYA对高糖诱导下RF/6A细胞有显著抑制作用,其可能机制为HSYA抑制血管内皮细胞VEGF表达。

Abstract:Objective To study the influence of Hydroxy Safflor Yellow A on the migration and VEGF expression of retinal and choroid endothelial cell in Rhesus monkey under high glucose condition.Methods We observed the influence of Hydroxy Safflor Yellow A on the migration and VEGF expression of retinal and choroid endothelial cells in Rhesus monkey under high glucose condition by cell scratch healing assay and half fix quantified reverse transcription-polymerase chain reaction (RT-PCR).Results Remarkable inhibition of the migration and VEGF expression of RF/6A with high glucose condition(22mmol/L) could be found when the concentration of HSYA was at 18 mg/L,37 mg/L,73mg/L.but the effect of HSYA was not powerful with normal glucose condition(5mmol/L).Conclusion HSYA may remarkably inhibit the migration and VEGF expression of RF/6A with high glucose,The mechanism may be related to the effect of HSYA on VEGF expression of RF/6A.
