| 摘 要： | 本实验以兔肌乳酸脱氢酶（ＬＤＨ）为受试对象，在ｐＨ７．０，２５℃，１０１３ｋＰａ条件下，采用不同配对的底物浓度（ＮＡＤＨ和丙酮酸）方式，测定了不同时间的光吸收值（Ａ３４０ｎｍ），并计算出ＮＡＤＨ在Ａ３４０ｎｍ每分钟减少的平均差值（△Ａ３４０ｎｍ／ｍｉｎ）。用二次作图法求出了ＬＤＨ的Ｋｍ（ＮＡＤＨ）值为１０６×１０－５ｍｏｌ／Ｌ，此结果与文献报道接近（１１×１０－５ｍｏｌ／Ｌ）。

| Abstract: | The enzyme reaction of LDH from reabbit muscle was carried out at pH 7.0,25,101.3kPa. Under different pairs of NADH and pyruvate as substrate,values of A340nm were determinated at different times.Average values of math.difference per min(A340nm/min) were calculated.Km(NADH)=1.0610-5mol/L was obtained by the plot of two steps.This Km(NADH) is approximative to that reported in the literature (1.110-5mol/L). |
