妊娠期糖尿病胎盘脂联素及其受体基因mRNA的表达

目的:探讨妊娠期糖尿病胎盘脂联素及其受体基因mRNA表达变化情况。方法:采用逆转录聚合酶链反应技术(RT-PCR)检测20例妊娠期糖尿病产妇、25例正常产妇胎盘脂联素及其受体基因mRNA的表达,并进行半定量分析。结果:①脂联素及其受体1/2基因mRNA在胎盘组织均有表达;②妊娠期糖尿病组胎盘脂联素基因mRNA表达水平(0.78±0.19)显著低于正常对照组(1.16±0.18),t=-6.76,P<0.001;妊娠期糖尿病组胎盘脂联素受体1基因mRNA的表达水平(2.04±0.72)与正常对照组(2.03±0.69)比较差异无统计学意义,t=0.06,P>0.05;妊娠期糖尿病组胎盘脂联素受体2基因mR-NA的表达水平(1.64±0.40)显著高于正常对照组(1.20±0.26),t=4.38,P<0.001。结论:胎盘部位有脂联素及其受体的表达,妊娠期糖尿病病理状态可影响脂联素及其受体2基因水平在胎盘的表达,但对受体1的表达无明显影响。

The study on the mRNA expression of adiponectin and its receptors in placenta of gestational diabetes mellitus

Objective:To study the variation of the mRNA expression of adiponectin and its receptors in placenta of gestational diabetes mellitus (GDM).Methods:The adiponectin and its receptors mRNA expression level in placenta of 20 mothers with GDM and 25 cases of normal control were evaluated by RT-PCR.Results:①The adiponectin and its receptors mRNA were expressed in placenta.②The expression of adiponectin mRNA in GDM group(0.78±0.19) was significantly lower than that in the control's(1.16±0.18,t=-6.76,P<0.001); The expression of adiponectin receptor-1 mRNA were no significant difference between GDM group of (2.04±0.72) and control group (2.03±0.69) (t=0.06,P>0.05) ; The expression of adiponectin receptor-2 mRNA in GDM group (1.64±0.40) was significantly higher than that in the control's (1.20±0.26)(t=4.38,P<0.001).Conclusion:There are expression of adiponectin and its receptors mRNA in placenta.The adiponectin and adiponectin receptor-2 mRNA expression level in placenta may be been influenced in GDM,but adiponectin receptor-1 mRNA wasn't.
