豚鼠近视眼视网膜Müller细胞的原代培养

摘    要:
目的研究豚鼠近视眼视网膜Müller细胞原代培养的方法。方法用半透明眼罩遮盖法建立豚鼠的近视眼模型。采用酶消化法培养豚鼠近视眼视网膜Müller细胞,用倒置相差显微镜观察Müller细胞的生长状况,以GFAP、Vimentin免疫组化染色进行细胞鉴定。结果半透明眼罩遮盖豚鼠右眼2周后,遮盖眼眼轴延长,近视形成。原代培养的视网膜Müller细胞贴壁生长,胞体较大、扁平,GFAP、Vimentin免疫组化染色呈阳性。结论酶消化法是培养豚鼠近视眼视网膜Müller细胞的一种有效方法。

Abstract:
Objective To investigate a technique for primary culture of retinal Müller cells in guinea pig myopia. Methods Guinea pig myopia was induced with translucent goggles. The retinal cells in guinea pig myopia were cultured using the enzyme-digesting method. Müller cells were identified by immunocytochemistry of GFAP and Vimentin, and cell growth status was observed with an inverse phase microscope. Results After two weeks, the right eye that had worn the translucent goggles became myopic with its ocular axis elongating. The retinal Müller cells attached to the walls of the dishes used in the primary cultures while growing and showed a large, flattened shape. These cells showed positive staining for GFAP and Vimentin. Conclusion The enzyme-digesting method is an effective method for obtaining retinal Müller cells in guinea pig myopia.
