摘    要:
目的:观察釉基质蛋白对人牙周膜细胞总蛋白含量及超微结构的影响。方法:组织块法培养人牙周膜细胞。考马斯亮蓝法测定人牙周膜细胞的总蛋白含量。透射电镜技术观察细胞超微结构。结果:釉基质蛋白质量浓度在50mg/L时即可明显增加人牙周膜细胞的总蛋白含量,l00mg/L时细胞的总蛋白含量增加到最大,此时,细胞的超微结构显示:核仁明显,粗面内质网及高尔基复合体发达。结论:釉基质蛋白能促进人牙周膜细胞核酸及蛋白质的合成活性。

Abstract:
AIM: To observe the effects of enamel matrix proteins (EMPs) on protein content and ultrastructureof human periodontal ligament cells(PDLC) in vitro. METHODS:The; fourth passage of PDLC was cultured in standard environment with or without EMPs for 5 days. Then the total protein content was measured by using Coomassie brilliant blue staining. After that, transmission electron microscope was used to observe the ultrastructure of PDLC cultured with the most optimal concentrations of EMPs. RESULTS; The significant promoting effect on protein synthesis was observed with 50 mg/L EMPs. The most optimal concentration of EMPs was 100 mg/L. In this condition, the quantity of rough endoplasmic reticulum, Golgi complex and nucleolus in PDLC changed higher. CONCLUSION: The results suggested that enamel matrix proteins could activate the periodontal ligament cells to make up proteins and nucleic acid.
