前列腺素E1脂微球载体制剂对大鼠急性肾缺血再灌注损伤的保护作用

摘    要:目的探讨前列腺素E1脂微球载体制剂(Lipo-PGE1)对大鼠肾脏缺血再灌注损伤的保护作用。方法采用在体大鼠肾脏缺血再灌注模型,以生理盐水空白对照观察前列腺素E1脂微球载体制剂对肾脏缺血再灌注损伤大鼠血浆中超过氧化物歧化酶(SOD)、丙二醛(MDA)、肌酐(Cr)、尿素氮(BUN)含量变化的影响,并取肾脏组织作病理检查。结果与模型对照组相比前列腺素E1脂微球载体制剂治疗组可提高缺血再灌注损伤大鼠血浆中SOD活力(194.31±8.02)u/Lvs(185.18±7.03)u/L;降低再灌注大鼠血浆中MDA含量(7.48±0.75)mmol/Lvs(8.22±0.57)mmol/L;明显改善肾功能(血Cr、BUN浓度分别是(112.77±12.29)mmol/L,(30.53±2.73)mmol/L及(127.35±14.78)mmol/L,(35.16±5.39)mmol/L;肾小管损伤病理分级显著减轻。结论前列腺素E1脂微球载体制剂对肾脏缺血再灌注损伤具有保护作用。

Abstract:To observe whether Lipo- PGE1 provide the protective effect on the ischemic-reperfusion renal injure. I/R lesions were produced in rats by clamping the renal arteries for 60min to induce warm ischemia and followed by reperfusion. Lipo-PGE1 was used at renal ischemia-reperfusion models while the control group was therapy with saline. The changes of plasmia superoxide dismutase (SOD), malondial-dehyde (MDA), blood ureanitrogen (BUN ) and serum creatinine(Cr) levels were measured. In addition, the histology of renals was evaluated after the ischemia-reperfusion injure. Compared to the control group, Lipo-PGE1 treated group had a significantly lower MDA, a significantly higher SOD,and a better renal function. The pathological grading of injured renal tubule were lower than those of control group. [Conclusions] Lipo-PGE1 does provide a certain degree of protection from ischemic-induced acute renal injure in rat.
