Expression of CCR3 in renal clear cell carcinoma and its effect on invasion and metastasis of renal clear cell carcinoma cells

Objective To investigate the expression of CCR chemokine receptor 3(CCR3)in renal clear cell carcinoma(RCC)and its effect on invasion and metastasis of RCC cells.Methods From April 2019 to April 2020,50 cases of renal clear cell carcinoma tissue specimens treated in the First Affiliated Hospital of Hunan College of Traditional Chinese Medicine were retrospectively selected,and 50 cases of adjacent tissues were selected as the control group for immunohistochemical staining.Human renal clear cell carcinoma cell line 786-0 and human renal clear cell carcinoma cell line A498 were selected for cell culture.CCR3 mRNA expression level was detected.CCR3 silencing lentiviral vector was constructed as the CCR3 inhibition group.A nonsense sequence was added to the control group,and physiological saline was added to the blank group.Cell migration and invasion capabilities were observed using cell migration and invasion experiments.The levels of phosphorylated extracellular regulated protein kinases(p-ERK1/2),matrix metalloproteinase-2(MMP-2),matrix metalloproteinase-9(MMP-9)were detected by Western blotting.Results The positive expression rate of CCR3 in renal clear cell carcinoma tissue(48.00%)was lower than that in adjacent tissues(12.00%),the difference was statistically significant(P<0.05).The expression of CCR3 mRNA in A498 cell line was higher than that in 786-0 cell line,the difference was statistically significant(P<0.05).Compared with the blank group,the number of cancer cell migration and invasion increased in the control group,while the number of cancer cell migration and invasion decreased in the CCR3 inhibition group;compared with the control group,the number of cancer cell migration and invasion decreased in the CCR3 inhibition group,the difference was statistically significant(P<0.05).Compared with the blank group,the expression of p-ERK1/2,MMP-2,MMP-9 protein in the control group increased,while the expression of p-ERK1/2,MMP-2,MMP-9 protein in the CCR3 inhibition group decreased;compared with the control group,the expression of p-ERK1/2,MMP-2,MMP-9 protein in the CCR3 inhibition group decreased,the difference was statistically significant(P<0.05).Conclusion CCR3 is highly expressed in renal clear cell carcinoma cells.Inhibition of CCR3 expression can inhibit the invasion and migration of renal clear cell carcinoma cells by regulating p-ERK1/2,MMP-2 and MMP-9 proteins.
