摘    要:目的:探讨纤维连接蛋白(FN)及透明质酸(HA)在高氧诱导新生鼠肺损伤发生中动态变化及意义。方法:将160只新生鼠随机分为实验组和对照组,每组80只,采用高浓度氧诱导的肺损伤模型,于实验后1,3,7,14,21d,观察支气管肺泡灌洗液(BALF)中嗜中性粒细胞及肺组织含水量的变化,并应用酶联免疫吸附法测定肺组织FN及HA的含量。结果:与对照组比较,在实验组肺组织含水量1d增加(P<0.05),3d达高峰(P<0.01),7d仍高于正常(P<0.05);BALF中嗜中性粒细胞的水平7,14,21d均明显升高(P<0.01);而FN含量3d开始升高(P<0.05),7d达高峰(P<0.01),14d仍高于正常(P<0.05);HA含量1d开始升高(P<0.05),3d达高峰(P<0.01),7d仍高于正常(P<0.05)。结论:暴露高氧环境中新生鼠的肺组织FN及HA水平增加可能与早期肺泡炎性反应和肺间质水肿密切相关。

Abstract:Objective:To investigate the dynamic change of fibronectin and hyaluronic acid in newborn rats with hyperoxia-induced lung injury and its significance. Methods:A total of 160 newborn rats were randomly and equally divided into experimental group and control group. The rat model of hyperoxia-induced lung injury was established in the experimental group. The changes of the number of neutrophils in bronchoalveolar lavage fluid (BALF) and the lung water content were observed,and the levels of fibronectin and hyaluronic acid were determined with enzyme-linked immunosorbent assay on days 1,3,7,14,and 21. Results:Compared with the control group,in experimental group,the lung water content,i.e. the lung dry to wet weight ratio,increased from day 1 (P < 0.05),reached the peak on day 3 (P < 0.01),and remained high on day 7. The number of neutrophils in BALF in experimental group was significantly higher than that in control group from day 7 to day 21 (P < 0.01). The level of fibronectin increased from day 3 (P < 0.05),reached the peak on day 7 (P < 0.01),and remained high on day 14 (P < 0.05). The level of hyaluronic acid increased from day 1 (P < 0.05),reached the peak on day 3 (P < 0.01),and remained high on day 7 (P < 0.05). Conclusion:The levels of fibronectin and hyaluronic acid increase in newborn rats exposed to hyperoxia,which may play an important role in the alveolar inflammation and interstitial edema.
