甘草与附子配伍对乌头碱、新乌头碱、次乌头碱大鼠药动学的影响

摘    要:目的:建立HPLC-MS同时测定大鼠血浆中的乌头碱、新乌头碱、次乌头碱含量的方法,并用于研究大鼠口服3种含附子煎液后乌头碱、新乌头碱、次乌头碱的药动学变化。方法:3组大鼠分别灌胃附子煎液(A液)、甘草附子单煎合并液(B液)、甘草附子合煎液(C液),附子剂量均为1.5 g/kg,HPLC-MS法测定乌头碱、新乌头碱、次乌头碱的血药浓度,用DAS 2.0数理统计软件分别计算其药动学参数。结果:甘草与附子配伍后,三种乌头碱的Cmax、AUC均降低,MRT、t1/2均延长,Tmax无明显变化。甘草附子合煎液相比于甘草附子单煎合并液,Cmax、AUC更低,MRT、t1/2更长。结论:甘草能够显著影响附子中三种乌头碱的药动学。
