PTEN蛋白在前列腺癌中的表达及意义

摘    要:目的 研究前列腺癌PTEN蛋白的表达及其与肿瘤转移、分级及分期的关系。方法 采用抗PTEN的多克隆抗体免疫组织化学技术染色法(Envision法)研究35例前列腺癌组织切片。结果 35例前列腺癌中有8例(22.9%)呈不同程度的阳性表达。转移者的PTEN表达率(5.9%)明显低于非转移者(38.9%,P<0.01);在A、B期的表达率明显高于C、D期(P<0.05);高、中分化组间表达率的差异虽无显著性,但高分化与低分化、中分化与低分化间PTEN阳性表达率的差异有显著性(P<0.05)。PTEN蛋白在良性组织上皮细胞及肿瘤细胞胞质中表达,在胞核及基质中不表达,在癌旁增生组织上皮细胞中呈强弱不等的阳性表达。结论 PTEN可作为良好的前列腺癌进展的肿瘤标记物。

Abstract:Objective To evaluate the significance of phosphatase and tensin homolog deleted chromosome ten (PTEN) in prostatic cancer(PCa). Methods The expression of PTEN was examined in tissue specimens resected from 35 patients with PCa by staining with a polyclonal antibody against PTEN. Correlations between the expression of PTEN and clinicopathologic factors were studied. PTEN was detected in 8 (22.9%) PCas. Results PTEN positivity was correlated well with metastasis and there were significant differences between stage A or B and C or D tumors. The differences were also significant between highly, moderately differentiated and poorly differentiatied cancers. Conclusion Immunohistochemical study revealed that PTEN was primarily localized in the cytoplasm of the benign prostatic hyperplasia (BPH) cells and cancers cells. PTEN can serve as a biomarker for PCa.
