强肌健力饮对脾虚大鼠性激素水平的影响

摘    要:目的:动态观察强肌健力饮对实验性脾虚证大鼠血清睾酮(T)、雌二醇(E2)、淀粉酶水平的影响及免疫器官组织形态学的改变,以期进一步了解该方药对改善脾虚证的意义及探讨中医脾肾互相关系的物质基础。方法:选用SD雄性大鼠,随机分为正常对照组,脾虚模型组,强肌健力饮高、中、低剂量组,四君子汤组。采用大黄复制脾虚证动物模型,分别给予蒸馏水、大黄、强肌健力饮(剂量分别为23.4g/kg、15.6g/kg、7.8g/kg)、四君子汤(20g/kg)灌胃。观察各组动物T、E2、淀粉酶水平的变化及脾脏、胸腺、肾上腺组织形态学的改变。结果:脾虚模型组大鼠造模10d时血清E2、T含量均比正常对照组显著升高(P〈0.05±0.01);造模20d后,脾虚模型组动物血清E2含量比正常对照组显著升高(P〈0.01),T含量则显著下降(P〈0.01)。灌服强肌健力饮10d时能显著降低E2、T含量(P〈0.01),而E2/T比值无改变;到给药20d时则能显著降低E2含量和E2/T比值(P〈0.05±0.01)。脾虚模型组大鼠造模10d和20d时血清淀粉酶含量均比正常对照组降低(P〈0.05),经灌服不同剂量强肌健力饮后,其血清淀粉酶含量改变不明显。脾虚模型组动物脾脏和胸腺质量及脏器指数均下降,组织结构均有明显破坏,肾上腺皮质也有不同程度的损伤,强肌健力饮能使已遭损伤的脏器组织得以修复,恢复到正常或接近正常水平。结论:脾虚证的发生与性激素水平紊乱及淀粉酶合成与分泌减少有关。而强肌健力饮能改善下丘脑-垂体-性腺轴功能,并对受损的脾脏、胸腺、肾上腺组织的修复具有显著的促进作用,从而发挥其健脾益气,强肌健力作用。提示T、E2这类物质可能是脾肾相关的重要物质基础之一,而脾脏、胸腺、肾上腺组织形态的改变可能是中医"脾虚及肾"病理形态学依据之一。

Abstract:Objective To investigate the therapewtic efficacy of Qiangji Jiardi Yin on male rat models of splenoasthenic syndrome through changes of serum sex hormones (T, E2 ), amylase and histologic changes of spleen, thymus, adrenals as well as to study the material foundation of spleno - renal mutual correlationship in traditional Chinese medicine. Methods Rat models male of splenoasthenic syndrome were established with daily gavage of rhubarb decoction (2ml 〈 200% ) for 20 days. These models were further divided into: (1) untreated group, n = 12 (2) treated with simultaneous daily gavage of Qiangji Jianli Yin at low (7.8g/kg, n = 13), moderate ( 15.6g/kg, n = 13) and high (23.4g/kg, n = 11 ) doses groups (3) treated with simultaneous daily garage of Si Jun Zi Tang (20g/kg, n = 10). Serum T, E2 and amylase levels were determined with RIA on d10 and d20 and the animals were sacrificed on d20 to procurespleen, thymus and adrenals for histologic stady. Control rats (n = 10) were given daily gavage of distilled water only. Results Serum E2 and T levels in the splenoasthenic syndrome models without treatment were significantly higher than those in controls rats on d10 (P 〈0.05 -0.01). On d20, the serum E2 levels increased further but T levels dropped markedly and were significantly lower than those in untreated group (P 〈0.01 ). The increases of E2, T on d10 were much less in the models treated with Qiangji Jianli Yin with maintainence of E2/T ratio. On d20 the serum E2 levels, though increased, were much lower than those in untreated group, bence the E2/T ratio was also much lower than that in untreated group and differed less from that in controls. Scrmn amylase levels on d10 and d20 in the sploneastheic models without treatment were significantly lower than those in controls rats (P 〈 0.05) and changed little after Qianji Jianli Yin administration.
