黄芪建中汤对慢性阻塞性肺疾病稳定期大鼠mtDNA ATPase6、ATPase8基因表达的影响

摘    要:目的 探讨黄芪建中汤对慢性阻塞性肺疾病(COPD)稳定期大鼠mtDNA ATPase6、ATPase 8基因表达的影响.方法 将120只SD大鼠随机分为6组:即A组(正常对照组)、B组(COPD模型组)、C组(黄芪建中汤高剂量组)、D组(黄芪建中汤中剂量组)、E组(黄芪建中汤低剂量组)、F组(金水宝胶囊组),每组20只.后五组采用气管内注射脂多糖(LPS)加烟熏的方法制作COPD大鼠模型,造模成功后,各治疗组分别给药8周,采用RT-PCR方法检测大鼠动脉血mtDNA ATPase6、ATPase 8基因表达变化.结果 A组的荧光生长曲线呈明显阳性;B组大鼠mtDNA ATPase6、ATPase8荧光反应较A组明显下降(P<0.01);与B组相比:C组、D组、E组、F组荧光反应明显升高(P<0.01),升高的次序为C组>D组>F组.C组与D组之间差异显著(P<0.05),D组与E组之间差异显著(P<0.05),C组、D组与F组之间差异显著(P<0.05).结论 模型组大鼠mtDNA ATPase6、ATPase8基因表达量减少,黄芪建中汤对mtDNA ATPase 6、ATPase 8的基因表达有明显的保护与恢复作用,疗效优于金水宝胶囊组.

Effect of Huangqi Jianzhong Decoction on mtDNA ATPase 6, ATPase 8 Gene Expression of Rats with Chronic Obstructive Pulmonary Disease in Stationary Phase

Abstract:Objective :To study the effect of Huangqi Jianzhong Decoction(HJD) on mtDNA ATPase 6, ATPase 8 gene expression of chronic obstructive pulmonary(COPD) in stationary phase. Methods: 120 SD rats were randomly divided into six groups, Group A( control group), Group B( COPD model group), Group C (HJD high -dose group), Group D (HJD medium -dose group), Group E (HJD medium low- dose group) ,Group F(Jin~huibao Capsule group) ,20 rats per group. The latter five groups were given with LPS intratracheal injec- tion plus smoked way to induce COPD rats. Each of the treatment group was administered for 8 weeks. RT - PCR method was used to de- tect the changes of the mtDNA ATPase 6,ATPase B gene expression of arterial blood of the rats. Resuhs:The fluorescence growth curve of Group A was significantly positive, the fluorescence response of the mtDNA ATPase 6 ,ATPase 8 of Group B was significantly lower than that of Group A(P 〈0. 01 ). Compared with Group B,the fluorescence response of Group C,D,E,F was significantly higher(P 〈0. 01 ) ,and their increase mxler was Group C 〉 Group D 〉 Group F. The difference between Group C and D was significant( P 〈 0. 05 ), the difference between Group D and E was significant( P 〈 0.05 ), the difference between Group C, D and E was significant( P 〈 0. 05 ). Conclusion :The gene expression of the mtDNA ATPase 6 ,ATPase 8 of the model rats decrease and Huaagqi Jianzhong Decoction could protect and restore the gene expression ans has a better effect than that of Jinshuibao capsule group.
