| 摘 要： | 目的研究细胞外信号调节激酶(ERK)通路与基质金属蛋白酶-9(MMP-9)在肺腺癌表达中的相关性及意义。方法应用免疫组织化学(SP)法检测磷酸化细胞外信号调节激酶(pERK)、MMP-9在67例肺腺癌组织中的表达。Western blot检测PD98059阻断ERK信号通路后肺癌细胞MMP-9蛋白表达水平的变化。结果 pERK蛋白在肺腺癌组织和正常组织中高表达率分别为65.6%、14.3%;MMP-9蛋白在肺腺癌组织和正常组织中高表达率分别为73.1%、21.4%。pERK与MMP-9表达存在正相关(r=0.57,P<0.05)。pERK、MMP-9的表达均与肺腺癌的TNM分期及淋巴转移状况相关(P<0.05),而与患者年龄、肿瘤大小无明显相关性(P>0.05)。用ERK阻断剂PD98059阻断ERK信号通路可降低MMP-9蛋白表达水平,并且随PD98059浓度升高MMP-9蛋白表达水平逐渐降低。结论 ERK信号通路可能通过上调MMP-9的表达促进肺腺癌的恶性进展,ERK信号通路可能是肺腺癌侵袭和转移的重要途径。 |
