蛇毒神经生长因子对大鼠视神经夹伤保护的电镜观察

摘    要:
目的研究蛇毒神经生长因子在视神经损伤后对视网膜神经节细胞的保护作用。方法将Wistar大鼠40只随机分为实验对照组和实验治疗组。制作实验性视神经夹伤模型,用头部宽1mm的微型血管夹夹伤大鼠右眼视神经后,实验治疗组向伤眼玻璃体腔内注入蛇毒神经生长因子100BU(0.025mL)。实验对照组向伤眼玻璃体腔内注入0.025mL平衡盐液。于损伤后第3d、7d、14d、30d、60d取材,用透射电镜观察不同时间段各组视网膜形态学变化。结果电镜下大鼠视网膜改变:实验治疗组和对照组电镜下均可见坏死和凋亡。伤后14d,实验治疗组视网膜微管数目比实验对照组较多,排列比较整齐。结论在视神经损伤早期,蛇毒神经生长因子能减轻视神经夹伤后微管的损坏,提高视网膜神经节细胞的存活数量,对视网膜神经节细胞有明显的保护作用。

Abstract:
Objective To study the neuroprotective effects of venom nerve growth factor(vNGF) on retinal ganglion cells after optic nerve crush in rat.Methods Forty Wistar rats were divided into experimental and experimental control groups at random.Using mini-vascular nip crushed the rats' optic nerves to make the crushed model.After crushed the optic nerve of the right eyes,100 BU(0.025 mL) of vNGF was injected into the vitreous of experimental group injury eyes.0.025mL of Balance salt solution was injected into the vitreous of experimental control group injury eyes.On the 3rd,7th,14th,30th and 60th days after injury,the changes of the retina were observed by using electron microscope.Results The changes of the retina were observed by using electron microscope:necrosis and apoptosis of RGCs in experimental and experimental control groups were seen under electron microscope.On the 14th days after injury,the numbers of microtubules in the treatment group were higher than those in the experimental control group obviously.Conclusion In the early stage of optic nerve injury,vNGF is effective in promoting the function of the optic nerve by reducing the injury of microtubules and increasing the number of survival retinal ganglion cells.
