蓝萼甲素对血管内皮细胞的保护作用

摘    要:目的:探讨蓝萼甲素对人脐静脉内皮细胞(HUVECs)的保护作用。方法:0.1 mmol/L过氧化氢(H2O2)作用于人脐静脉内皮细胞株,实验分为空白对照组、氧化损伤模型组、阿司匹林对照组和蓝萼甲素低、中、高剂量组。RT-PCR检测内皮细胞ET-1、iNOS、eNOS的表达。结果:与模型组比较,蓝萼甲素低、中、高剂量组均能升高细胞增殖率,蓝萼甲素中、高剂量组ET-1和iNOS表达明显减少,eNOS表达明显增多。结论:蓝萼甲素能够明显改善H2O2对内皮细胞的损伤,可能是通过降低ET-1而减少血管的收缩;降低iNOS表达而减少内皮细胞的损伤,达到保护内皮细胞的作用。

Abstract:Objective To study the protective effects of glaucocalyxin A on HUVEC cells. Methods: HUVEC cell injurymodel was induced by H202, and there were blank control group, Oxidative injury model group, aspirin control group andglaucocalyxin A low, middle and high dose group; The expression of ET-1, iNOS and eNOS in molecular level were detectedwith RT-PCR. Results: Compared with the model group, Glaucocalyxin A low, middle and high dose group could significantlyincrease cell proliferation rate; ET-1 and iNOS expression decreased and eNOS expression markedly increased, in glaucocalyx-in A middle and high dose group. Conclusion: Glaucocalyxin A can protect endothelial cells damage due to H202, which mayrelated to reduction of constriction of the blood vessels by reduce ET-1, and reduce endothelial cell damage according to de-cline of iNOS expression, to protect endothelial cells.
