摘    要:
目的在前期实验获得的小细胞肺癌相关基因片段L2基础上进行该基因的验证及cDNA末端快速扩增法(RACE)扩增。方法根据小细胞肺癌相关基因片段L2的已知序列,设计引物,提取小细胞肺癌细胞株(NCI-H446)的RNA并行反转录-聚合酶链反应(RT-PCR)。在获得的阳性片段的基础上进行RACE扩增并测序,将测序结果与Genbank序列进行对比。结果通过RT-PCR获得小细胞肺癌相关基因片段L2,且与目的片段相近,将其进行RACE扩增后与Genbank序列进行对比其序列与ABCE1的序列基本一致。结论通过小细胞肺癌RT-PCR及RACE扩增得到了ABCE1基因,该基因可能与小细胞肺癌存在一定的关系。

Abstract:
Objective To verify a novel small lung cancer related gene fragment L2 which was gained by earlier trail and RACE-amplifed positive fragment.Methods On the basis of the alignment of small lung cancer related gene fragment L2,a pair of primiers was designed,RNA was extracted from small lung cancer(NCI-H446),CDNA fragment was gained by RT-PCR.To obtain positive fragment and then RACE perfrmed.Checked up the full length then compairied with Genbank.Results L2 fragment was obtained from NCI-H446.After RACE amplified,the full length alignment was correspondent with ABCE1 gene.Conclusion Through RT-PCR and amplification,ABCE1 gene was obtained.This gene may be related with small lung cancer.
