反相高效液相色谱法同时测定脉络宁注射液中6种酚酸类成分的含量

摘    要:目的 建立用反相高效液相色谱法同时测定脉络宁注射液中6种酚酸含量的方法。方法采用Kromasil-C18柱(4.6mm×250mm,5μm);流动相:乙腈-0.1%磷酸溶液,梯度洗脱;流速:1.0mL/min;紫外检测波长:327nm;柱温:25℃;进样量:20gL。结果新绿原酸、绿原酸、隐绿原酸、咖啡酸、3,5-二咖啡酰奎宁酸、3,4-二咖啡酰奎宁酸的线性范围分别为0.01090~0.5448μg(r=0.9999)、0.02406~1.2032μg(r=1)、0.01106~0.5528μg(r=1)、0.01594~0.7968μg(r=1)、0.009104~0.4552μg(r=1)、0.01037~0.5184μg(r=1),回收率分别为101.06%、100.22%、101.54%、99.42%、100.21%、101.65%。结论 本法灵敏简便,准确可靠,可作为脉络宁注射液中新绿原酸、绿原酸、隐绿原酸、咖啡酸、3,5-二酰奎宁酸、3,4-二咖啡酰奎宁酸含量同时测定的方法。

Determination of Six Phendic Acids in Mailuoning Injection by RP-HPLC

Abstract:Objective To establish a method for determination the contents of six phendic acids in Mailuoning injection by HPLC at same time. Methods Separation was performed on a Kromasil-C18 (4.6 mm× 250 mm, 5 μm) column with mobile phase consisting of 0.1% phosphoric acid-water and Acetonitrile with gradient elute at the flow rate of 1 mL/min. The UV detection wavelength was set at 327 nm and the column temperature was set at 25℃. The sample size was 20 μL. Results The standard cures of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, caffeic acid, 3,5-Dicaffeoylquinic acid, 3,4- Dicaffeoylquinic acid were linear within the contentration range of 0.010 90-0.544 8μg (r =0.999 9), 0.02406-1.2032 μg (r= 1), 0.01106-0.552 8 μg (r= 1), 0.01594-0.7968 μg (r= 1), 0.009 104-0.4552 μg (r = 1), 0.010 37-0.518 4 μg (r = 1) respectively. And the recovery rates of them were 101.06%, 100.22%, 101.54%, 99.42%, 100.21%, 101.65% respectively. Conclusion The method appeared to be simple, reliable, accurate and can be used to determine the contents of six phendic acids in Mailuoning injection.
