Caspase阻滞剂zVAD-fmk对大鼠脊髓损伤细胞凋亡的影响

摘    要:目的:观察Caspase阻滞剂zVAD-fmk对大鼠脊髓损伤细胞凋亡的影响。方法:将48只大鼠制备为压迫性脊髓损伤模型。分为单纯损伤组24只及实验组(应用zVAD-fmk)24只。损伤后8h、3及7d对脊髓损伤区进行HE、细胞凋亡的检测,免疫组化检测Caspase-3的表达变化。同时采用BBB评分和斜板试验观察脊髓神经功能恢复情况。结果:2组均见TUNEL阳性的凋亡细胞和Caspase-3表达。实验组凋亡的细胞数减少。免疫组化检测Caspase-3表达降低,神经功能增强。结论:Caspase阻滞剂zVAD-fmk能减少继发性脊髓损伤细胞凋亡和促进神经功能的恢复。

Abstract:Objective: To investigate the effect of caspase inhibitor zVAD-fmk on the cell apoptosis following acute spinal cord injury in rats. Methods: The models of acute compression spinal cord injury in 48 female Sprague-Dawley rats were made. The 48 rats were randomly divided into 2 groups: control group (n=24) and experimental group (n=24, receiving zVAD-fmk). The animals were then sacrificed at 8 h, 3 d or 7 d after injury. The lesion areas of the spinal cord were cut for morphological studies by hematoxylin and eosin staining. Cell apoptosis was detected by Hoechst33342 staining and TUNEL method and the expression of Caspase-3 by immunohistochemistry. The neurofunction of spinal cord was measured by BBB score and platform test. Results: The apoptostic cells positive for TUNEL and Caspas-3 expression were detectable in both two groups. In the experimental group, the number of apoptotic cells and the expression of Caspase-3 were decreased and the neurofunction of the spinal cord improved as compared with those in the control group. Conclusion: Caspase inhibitor z-VAD-fmk can reduce the numbers of apoptotic cells and promote the nerve function recovery after spinal cord injury.
