摘    要:目的利用siRNA沉默子宫内膜癌lshikawa细胞中α-catulin基因的表达,探讨其对子宫内膜癌细胞增殖、凋亡与侵袭的影响。方法将培养的Ishikawa细胞分为pGCsi-α-catulin组、pGCsi-NC组与空白对照组(未加入任何试剂的细胞组)。构建α-catulin siRNA的表达载体,并转染Ishikawa细胞。Western blot方法检测转染前后Ishikawa细胞中α-catulin蛋白的表达;MTT实验检测细胞生长抑制率,流式细胞技术分析转染前后Ishikawa细胞的凋亡变化,Transwell实验检测转染前后Ishikawa细胞侵袭能力的变化。结果与pGCsi-NC组与空白对照组相比,pGCsi-α-catulin组Ishikawa细胞中α-catulin蛋白表达水平显著降低,Ishikawa细胞的增殖能力明显降低,凋亡率明显升高,侵袭能力明显减弱(P<0.05)。结论α-catulin可成为治疗子宫内膜癌细胞的靶点。

Abstract:Objective To investigate the effect of silencing alpha-catulin gene on the proliferation,apoptosis and invasion of endometrial cancer cells.Methods Ishikawa cells were divided into pGCsi-a-catulin group,pGCsi-NC group and blank control group(cells not treated with any reagent).The expression vector of alpha-catulin siRNA was constructed and transfected into Ishikawa cells.Western blot was used to detect the expression of alpha-catulin gene and protein in Ishikawa cells,MTT assay was used to detect the growth inhibition rate,flow cytometry was used to analyze the apoptotic changes of Ishikawa cells,and Transwell assay was used to detect the changes of invasive ability of Ishikawa cells.Results Compared with pGCsi-NC group or blank control group,the expression level of alpha-catulin was decreased significantly,the proliferation ability of Ishikawa cells was significantly decreased,the apoptotic rate was significantly increased,and the invasive ability was significantly weakened in pGCsi-a-catulin group.Conclusion Alpha-catulin could be a potential target in the treatment of endometrial cancer cells.
