乌龙丹对多梗塞性痴呆模型大鼠脑组织20种氨基酸含量的影响

摘    要:目的:改进多梗塞性痴呆(MID)大鼠模型的制作方法,并探讨乌龙丹对多梗塞性痴呆模型鼠全脑20种氨基酸含量的影响。方法:从颈内动脉推注干血悬液造模,以TTC病理染色法、迷宫实验对模型的成立进行验证,AA的测定采用反相高效液相色谱法,乌龙丹的制作采用水煮醇提法。结果:造模组TTC染色可见明显的白色缺血区,迷宫实验造模组大鼠躲避电击的错误次数明显多于正常组,而乌龙丹能减少造模大鼠的错误次数,且能提高痴呆晚期兴奋性AA的含量,降低其他AA的含量。结论:本MID方法造模简便,存活率成功率高;乌龙丹能改善模型大鼠的学习记忆功能,其作用机制可能与提高兴奋性AA的含量,降低抑制性AA的含量有关。

Abstract:Objective: To improve the model of MID in rats and explore the effect of 20 AA's content in brain on them treated with WLD. Methods: The model is made by injecting dry blood liquid from internal carotid artery and tested by triphenyl tetrazolium chloride (TTC) pathological dying、maze experiments. The contents of AA were measured by HPLC. The herbs of WLD were boiled with water, and extracted with alcohol. Results: Model rats have white area of ischemia in brain by TTC dying and make more wrong than normal rats when escape electrical shock. Error is decreased, the content of EAA raised and IAA dropped for the team of WLD. Conclusion: The methods of establishing model is more convenient. The survival and successtive rate is larger. The study and the memory are improved for the cats treated with WLD. The reason maybe connected with EAA's increment and IAA's reduction.
