摘    要:SNAFL-calcein-AM、rhodamine123分别标记培养的小鼠腹腔巨噬细胞胞浆pH、线粒体pH和线粒体跨膜电位;用激光共聚焦显微镜,观察了巨噬细胞内线粒体的分布,ConA刺激后胞浆pH、线粒体pH及线粒体跨膜电位随时间变化曲线。结果显示ConA刺激可引起线粒体内外pH差增加,线粒体跨膜电位在刺激开始时上升,而后逐渐下降;线粒体质子动力势绝对值在刺激开始一段时间增加,随后逐渐降低到比刺激前更低的水平。

Abstract:Cultured mouse peritoneal macrophage cytosolic pH,mitochondrial pH and transmembrane potential were labeled with SNAFL calcein,rhodamine 123,respectively.The changes of mitochondrial distributions in macrophage were observed and the kinetic changes of cytosoli pH,mitochondrial pH and transmembrane potential were given by a Laser Scanning Confocal Microscope after stimulated by Concanavalin A. These results showed Con A induced decrease of cytosolic pH,a little of change of mitochondrial pH, and mithondrial transmembrane rose at first,then gradually decreased.The power of mitochondrial synthesized ATP (proton motive force)rose at the begin,and following gradually decreased to the level which was lower than that of before stimulated by Concanavalin A.
