PTEN在食管鳞状细胞癌中的表达和意义

目的研究人第10号染色体缺失的磷酸酶及张力蛋白同源基因(PTEN)在食管鳞状细胞癌中的表达与临床病理特点的关系。方法采用免疫组织化学及荧光定量聚合酶链反应(PCR)法分别检测60例胸外科食管鳞状细胞癌手术患者食管鳞状细胞癌组织、癌旁组织和正常食管黏膜组织中PTEN的表达水平。分析PTEN在食管鳞状细胞癌浸润、分化、转移中的作用。结果 PTEN在正常食管组织(2.01±0.07)、癌旁组织(1.50±0.10)、食管鳞状细胞癌组织(0.89±0.08)中的表达逐渐降低(P0.01)。PTEN在有淋巴结转移的食管鳞状细胞癌组织中(0.87±0.08)的表达比在无淋巴结转移(0.94±0.07)的表达低;PTEN在高分化(0.91±0.06)、中分化(0.90±0.07)、低分化(0.85±0.10)的食管鳞状细胞癌组织中的表达逐渐降低,PTEN在浸润深(0.88±0.08)的食管鳞状细胞癌组织中的表达比浸润浅(0.93±0.07)的表达低(P0.05或0.01)。在60岁组与≥60岁组中的阳性表达差异无统计学意义(P0.05)。结论 PTEN基因的丢失或突变均与食管鳞状细胞癌的生长、分化和浸润转移相关。

Expression and significances of PTEN in esophageal squamous cell carcinoma

Objective To explore the expression of phosphatase and tensin homolog deleted on chromosome ten （PTEN） in the esophagus squamous cell carcinoma and its relationship with pathological features.Methods Immunohistochemistry and realtime fluorescence quantitative polymerase chain reaction（PCR）method were used to detect the expression level of PTEN in 60 cases of esophagus squamous cell carcinoma,adjacent tissue and normal esophageal mucosa.The difference and correlation of PTEN expression in infiltration,division,metastasis of esophagus squamous cell carcinoma were analyzed.Results The expression of PTEN was reduced in the normal mucosa （2.01± 0.07）,adjacent tissue （1.50 ± 0.10）and esophagus squamous cell carcinoma （0.89 ± 0.08）（P 〈0.01）.The expressionat of PTEN in esophageal squamous cell carcinoma with lymph node metastasis（0.87 ± 0.08）,was lower than that in esophageal squamous cell carcinoma without lymph node metastasis （0.94 ± 0.07 ）.The expression of PTEN was reduced in well-differentiated （0.91 ± 0.06 ）,moderately differentiated （0.90 ± 0.07 ） and poorly differentiated （0.85±0.10）of esophagus squamous cell carcinoma.The expression of PTEN in esophageal squamous cell carcinoma of deep infiltration （0.88 ± 0.08）was lower than that in esophageal carcinoma of shallow infiltration （0.93±0.07）（P〈0.05 or 〈0.01）.There was no statistic significance for PTEN expression between the expression rate of 〈60 and ≥60 years age group（P〉0.05）.Conclusion The loss or mutation of PTEN gene is correlated with the growth,differentiation,infiltration and metastasis of esophageal squamous cell carcinoma.
