电刺激对大鼠坐骨神经切断后脊髓运动神经元凋亡调控基因表达的影响

摘    要:
目的 观察直流电场对鼠坐骨神经切断后脊髓运动神经元bcl-2,bax基因表达的作用,探讨电刺激防治细胞凋亡的机制。方法 30只SD大鼠坐骨神经切断后,平均分为两组。实验组在相应的脊髓节段立即置入直流电刺激器,对照组置入无输出电流的刺激器,分别于术后1,4,7,14,28d取材,应用免疫组化技术,光镜下观察脊髓动物神经元bcl-2,bax的表达,并对脊髓阳性运动神经元进行计数。结果 坐骨神经切断后4,7,14d,实验组脊髓运动神经元bcl-2阳性数高于对照组;坐骨神经切断后4,7,14,28d实验组脊髓运动神经元bax阳性数低于对照组。结论 直流电场对鼠坐骨神经切断后脊髓运动神经元bcl-2,bax的表达具有调节作用。

Abstract:
Objective To observe the effect of direct-current field stimulation on bcl-2 and bax gene expression in spinal cord motor neurous after operation and to study the mechanism of intervention effect of electric stimulation on apoptosis. Methods In experimental group direct-current stimulators were put into the relevant spinal cord segments and electric stimulation were given instantly after sciatic nerve transection. In control group the same stimulators were put inside without current output. The specimens were taken at 1, 4, 7, 14, 28 days after operation and studied with immunohistochemistry method. The bcl-2 and bax gene expression were observed with light microscope. The spinal cord motor neurons with gene expressed were counted. Results At 4, 7, 14 days after operation in experimental group the number of spinal cord motor neurons with gene bcl-2 expressed was more than that in the control group, while the bax expressed motor neurons were less than those in the control group at 4,7, 14, and 28 days after operation. Conclusions Direct-current field may regulate the expression of the bcl-2 and bax gene after sciatic nerve transection in rats.
