摘 要: | 目的探讨人脑透明质酸结合蛋白(hbHABP)对人肿瘤细胞凋亡的影响。方法将hbHABP全长cDNA转染到人乳腺癌细胞(MDA435)和前列腺癌细胞(TSU)中,采用Western blot检测细胞caspase-3、CD95表达的变化,并采用酶联免疫吸附试验试剂盒测定细胞裂解液中Bcl-2、Cip/WAFI的含量。再将转hbHABP基因的MDA435细胞种植到鸡胚绒毛尿囊膜(CAM)上,使MDA435肿瘤细胞在体内环境中生长成瘤,取肿瘤块作病理切片,采用caspase-3抗体作免疫组织化学染色。结果MDA435和TSU细胞转染hbHABP基因后,caspase-3和CD95表达均增高;MDA435-hbHABP细胞裂解液中Bcl-2含量下降,TSU-hbHABP的Bcl-2无明显变化。TSU-hbHABP细胞Cipl/wAFl的含量升高,MDA435-hbHABP无变化。免疫组织化学结果显示,MDA435-hbHABP肿瘤组织内caspase-3表达均增高。结论hbHABP可能诱导肿瘤细胞凋亡,从而抑制肿瘤生长。

Experimental study of hyaluronic acid binding protein on human cancer cells apoptosis |

Abstract: | Objective To study the effects of human brain hyaluronic acid binding protein (hbHABP) on some human cancer cells apoptosis. Methods Full length cDNA of hbHABP was transfected into human breast cancer cell line (MDA435) and prostate cancer cell line (TSU). Caspase-3 and CD95 from these transfectants were detected by Western blot. Bcl-2 and Cip1/WAF1 contents from lysis of transfectants were determined by ELISA method. MDA435-hbHABP cells were implanted onto chorioallantoic membrane (CAM), and the pathologic section of the tumor was stained for caspase-3 expression. Results Expressions of caspase-3 and CD95 from both MDA435-hbHABP and TSU-hbHABP were increased. The Bcl-2 content from MDA435-hbHABP transfectants decreased (no change in TSU-hbHABP cells), and the Cip1/WAF1 from TSU-hbHABP increased (also no change in MDA435-hbHABP cells).Immunohistochemical staining showed that caspase-3 expression was much higher in MDA435-hbHABP tissues. Conclusion Human brain hyaluronic acid binding protein (hbHABP) may induce apoptosis in some human cancer cells in vivo and in vitro;and in term inhibit the growth of tumor. |
