肺结节病患者肺泡灌洗液中基因表达情况研究

摘    要:
目的了解肺结节病患者肺泡灌洗液中部分基因的表达情况。方法应用基因芯片技术检测3例肺结节病患者和1例对照者肺泡灌洗液中的基因表达,选择明显差异表达的基因进行生物信息学分析。结果肺结节病患者和对照者肺泡灌洗液中的基因表达存在明显差异,其中RGS5,CCL16,CCL5,IL-18的Cy5/Cy3比值〉2.0,基因表达上调;ADAM9和IL-6的Cy5/Cy3比值〈0.5,基因表达下调。结论CCL16、ADAM9、CCL5、IL-18、IL-6和RGS5与肺结节病的发生密切相关,深入研究这些基因的功能可进一步认识结节病的发病机制。

Abstract:
Objective To evaluate the gene expression in bronchoalveolar lavage fluid from patients with pulmonary sarcoidosis.Methods A cDNA microarray was used to detect the gene expression in bronchoalveolar lavage fluid from 3 patients with pulmonary sarcoidosis and 1 healthy person as a normal control.And the genes that express significantly different were chosen and analyzed with bioinformatics.Results Significant difference in gene expression between patients with pulmonary sarcoidosis and healthy person was found.Of which the radios of Cy5/Cy3>2.0 in RGS5,CCL16,CCL5 and IL18,the gene expression was up-regulated;but the radios of Cy5/Cy3<0.5 in ADAM9 and IL6,the gene expression was down-regulated.Conclusion Our findings suggest that CCL16,ADAM9,CCL5,IL18,IL6 and RGS5 are closely related to generation of pulmonary sarcoidosis.So deep study on the functions of these genes may further understand the pathogenesis of sarcoidosis.
