摘    要:
目的:探讨高碘对体外培养的人甲状腺细胞超微结构的影响与相关甲状腺疾病的关系。方法:采用体外甲腺细胞原代培养技术,术中取甲状腺腺瘤旁组织,常规培养7天后,分别加入不同浓度的KI溶液继续培养7天,收集细胞做透射电镜观察,结果:当碘浓度为0.006ug/ml,0.03ug/ml,0.15u/ml时,其细胞的超微结构无明显变化,当碘浓度为0.75ug/ml时,细胞出现微绒毛增加,核变形,核膜册陷,核二增大,双核仁,线粒体和粗面内质网增加,当碘浓度达到3.75ug/ml时,出现线粒体肿胀,粗面内持网扩张,当碘浓度为18.75ug/ml时,大部分细胞溶解,残余的细胞出现层状小体,结论:高碘可使甲腺细胞增殖活跃,功能增强,当碘浓度进一步增高时,。则出现细胞损伤,继而出现功能抑制,说明高碘在甲腺机能亢进症,甲腺炎以及甲腺癌的发病过程中起重要作用。

Abstract:
[Purpose] To investigate the high iodine effects on thyroid follicular cells by primary culture and the relationship between the iodine and thyroid diseases. [ Method ] Follicular cells were cultured for 7 days in different iodine concentrations medium, and then for the transmission electron microscopy (TEM). [ Results ]TEM study showed that there were no marked changes in ultrastructure at 0.006μg/ml, 0.03μg/ml, 0.15μg/ml iodine, with increasing in number of the microvilli, rough-surfaced endoplasmic reticulum (RER) and mitochondria were found with the 0.75μg/mliodine, as well as appearing nuclear defornity, enlarged nucleoli and binucleoli in the same iodine concentration. When the iodine concentration was 3.75μg/rnl, it appeared mitochondria swelling and RER expanded. Cell lysis could be seen with the 18.75μg/rnl iodine. [ Conclusion ] Thyroid follicular cells in high iodine concentration appear active proliferation, hyperfunction and thyrolytic. So the high iodine concentration played an important role in the genesis of hyperthyroidosis, thyroiditis and thyroid carcinomas.
