摘    要:目的 探讨富含血小板的血浆(PRP)对骨髓基质细胞DNA合成和细胞周期变化的影响。方法 以贴壁筛选法分离、培养狗的骨髓基质细胞,应用流式细胞仪观察PRP对骨髓基质细胞DNA合成和细胞周期变化的影响;通过透射电镜观察细胞的超微结构改变。结果 PRP对培养48小时的骨髓基质细胞S期的DNA含量明显增高,并促使细胞内线粒体更丰富,粗面内质网增多。结论 PRP可通过刺激骨髓基质细胞的DNA合成,促进其分裂、增殖及细胞的合成分泌。

Abstract:Objective To explore the effects of platelet-rich plasma (PRP) on the DNA synthesis of bone marrow stromal cells (BMSCs) and the changes of cellular period. Methods Cultured BMSCs in vitro were isolated and the effects of PRP on cellular DNA synthesis were observed by flow cytometer. The changes of BMSCs surface structure and ultrastructure were observed by electron microscope. Results PRP increased the DNA synthesis of cellular S period by promoting BMSCs from G0/G1 period into S period. The effects of PRP on increasing cellular DNA contents were most significant at 48 h culture. PRP increased BMSCs surface granules mitochondria and reticular fibers. Conclusion The results show that PRP could stimulate the BMSCs DNA synthesis and promote cell proliferation,fissiparism, and secretion in vitro.
