复方顺铂多相脂质体的初步研究

摘    要:
本文报道了制备多相脂质体复方顺铂(102)注射液的初步研究,对注入-超声法和成膜-超声法两种制剂工艺进行了比较,抑瘤试验表明,对小鼠腹注提高剂量后,(102)和顺氯氨铂(101)对S_(180)抑瘤率都较游离顺铂高,其中(102)对S_(180)抑瘤率稍高于(101),(102)的LD_(50)0(腹注、小鼠)26.52mg/kg,而游离顺氯氨铂的LD_(50) 12.45mg/kg。

Abstract:
In this paper, a study on the preparation of the suspension of polyphaseliposome cisplatin-co(102) is reported. Cisplatin-co (102) is composed of cisplatin, oleic acid, soybean lecithin,leucitin cholesterol, exeipients(Tween-80 and PVP etc.), 101 being without oleicacid, but similar to remainders of 102. A comparsion was made between theinjection-ultrasonic method and the method of membrane-forming-ultrasonic, whichwere used as in preparing 102 and 101. Preliminary results showed that 102 was stronger inhibitor against sarcomaS_(180) than 101, when intraperitoneally injected in mice. The new dosage form forantitumor agent 102 might be used clinically due to its higher antitumor activity,and lower toxicity. But further studies on it will be needed.
