bcl-2核酶对SMMC7721细胞端粒酶活性的影响作用

摘    要:目的 观察bcl-2核酶对SMMC-7721细胞的作用,探讨bcl-2表达水平的改变对细胞端粒酶活性的影响。方法 经脂质体介导的方法将PMTr-neo(下向bcl-2核酶真核表达载体)导入SMMC7721细胞中,细胞克隆转移扩大培养后,采用TUNEL,TRAP-PCR-ELISA,免疫组化技术结合图像分析技术检测SMMC7721/PMTr-neo细胞凋亡=细胞端粒酶活性及P16,P12的改变。结果 较对照组SMMC7721/PMTr-neo细胞Bcl-2表达水平显著下降,伴有显著细胞凋亡现象;同时端粒酶活性下降,P16,P21表达水平显著增加。结论 bcl-2核酶可促进SMMC7721细胞发生凋亡,并降低细胞端粒酶活性,提示Bcl-2表达水平的改变对细胞端粒酶活性有一定的影响。

Abstract:AIM To observe the effect of bcl 2 ribozyme on SMMC7721 cells and to investigate bcl 2 expression on cellular apoptosis and telomerase activity. METHODS PMTr neo (sense bcl 2 ribozyme eucaryotic expression vector) was introduced into SMMC7221 cells by using lipofectin media tion, after cloning TUNEL and TRAP in combination of ELISA and IHC in combination of imaging analysis was used to determine SMMC7721/PMTr neo apoptosis and telomerase activity. RESULTS Compared with control, SMMC7721/PMTr neo presented a significant drop in bcl 2 expression and an increase in P16 and P21 expression, accompanied by a decline in remarkable apoptosis and telomerase activity. CONCLUSION bcl 2 ribozyme promotes apoptosis of SMMC7721 cells and reduces telomerase activity. These provide a theoretical basis for applying antisense technique to treatment of hepatocarcinoma.
