表皮葡萄球菌对氟喹诺酮类药物摄入的研究

摘    要:
目的探讨耐药表皮葡萄球菌中是否存在主动外排系统,以揭示表皮葡萄球菌对氟喹诺酮类药物的耐药机制。方法应用荧光法检测表皮葡萄球菌耐药株和敏感株对环丙沙星、司帕沙星的累积量以及加入利血平和碳酰氰间氯苯腙(CCCP)后对环丙沙星、司帕沙星累积量的变化情况。结果表皮葡萄球菌临床株和敏感株中加入CCCP后菌体内环丙沙星、司帕沙星的累积量变化不大.而利血平可以使SR79菌体内环丙沙星的累积量显上升。结论表皮葡萄球菌中可能存在针对亲水性氟喹诺酮药物的主动外排系统。

Abstract:
Objective To explore whether active efflux system is present in Staphylococcus epidermidis in order to identify the resistance mechanism of S.epidermidis to antibiotics.Methods The accumulation of ciprofloxacin and sparfloxacin in S.epidermidis were measured by a fluorescence method and the influence of carbonyl cyanide m-chlorophenylhydrazone (CCCP) or reserpine was analyzed.Results Reserpine could dramatically increase the accumulation of ciprofloxacin in S.epidermidis SR79. However, CCCP had little effect on the accumulation of ciprofloxacin and sparfloxacin in clinical susceptible S. epidermidis isolates. Conclusions There may have an active efflux system targeting hydrophilic fluoroquinolones in S.epidermidis.
