地塞米松和辛伐他汀对成骨细胞增殖分化的影响

目的:研究地塞米松和辛伐他汀对成骨细胞增殖分化的影响.方法:从新生SD大鼠颅盖骨分离培养成骨细胞,随机分为3组培养,A组加入1×10-7 mol/L的地塞米松、1×10-7 mol/L的辛伐他汀和二甲基亚砜(DMSO,终浓度为1 g/L);B组加入1×10-7 mol/L的地塞米松和与A组等量的DMSO;C组只加入与A组等量的DMSO作为对照.96 h后,以RT-PCR一步法分析成骨细胞骨钙素mRNA的表达;采用MTT法测定成骨细胞增殖率.结果:A、C组成骨细胞骨钙素mRNA表达量及细胞增殖率均高于B组(P＜0.01),A与C组比较差异无统计学意义(P＞0.05).结论:辛伐他汀可对抗地塞米松对成骨细胞增殖和分化的抑制作用.

Effect of dexamethasone and simvastatin on the osteoblast proliferation and differentiation

Aim: To study the effect of dexamethasone and simvastatin on the osteoblast proliferation and differentiation. Methods: Osteoblasts were separated from skull of newborn SD rats, and randomly divided into 3 groups (8 samples for each group). Group A was fed with culture medium RPMI 1640,and treated with simvastatin of 10 -7 mol/L, dexamethasone of 10 -7 mol/L and DMSO of 0.1%;Group B was maintained in culture medium RPMI 1640 and treated with dexamethasone of 10 -7 mol/L and DMSO of 0.1%;Group C was only cultured with DMSO of 0.1%. After 96 hours, the expression of osteocalcin mRNA was observed by RT-PCR and the proliferation of osteoblast was measured using MTT reduction assay. Results: The expression of osteocalcin mRNA and the rate of proliferation of osteoblast of Group A and C were significantly higher than those of group B (P<0.01), and there was no significant difference between group A and group C (P>0.05). Conclusion: Simvastatin is resistant to the restraint of the osteoblast proliferation and differentiation by dexamethasone.
