红细胞变形性与心脏移植急性排斥反应的关系

摘    要:目的研究红细胞变形性的改变与心脏移植急性排斥反应的关系。方法随机选取6只SD大鼠作为正常对照组,采集正常红细胞变形指数。手术创伤组(A组,SD大鼠24只);同系移植组(B组,供受体均为SD大鼠,共48只);同种移植组(C组,供体为Wistar大鼠,受体为SD大鼠,各24只)。B组、C组应用ONO大鼠腹腔异位心脏移植模型。观察术后1、3、5和7d的移植心病理改变,同时采用激光衍射法测定红细胞变形指数。结果A组与B组之间差异无显著性(P>0.05),两组各观察点红细胞变形指数与正常组比较差异均无显著性(P>0.05)。C组红细胞变形指数术后第1天起持续下降,并在第5天达到变化的高峰,与正常组相比差异有显著性(P<0.05);与A组、B组比较差异均有显著性(P<0.001)。红细胞变形指数与心脏病理的相关分析显示两者具有显著的相关性(P<0.001,r=0.78)。结论红细胞变形性的下降与移植心脏急性排斥反应显著相关,可能是参与及促进急性排斥反应发生发展的重要因素之一。

Relationship between erythrocyte deformation and acute rejection after cardiac transplantation in rats

Abstract:To investigate the relationship between erythrocyte deformation and acute rejection after cardiac transplantation. 6 healthy male SD rats were drew randomly as normal group. 120 rats were randomly divided into surgery trauma group(group A, 24 SD rats), isograft group (group B, 48 SD rats) and allograft group (group C, 24 SD rats and 24 Wistar rats, consisted of Wistar donors into SD recipients). The model of rat heterotopic abdominal heart transplantation was used. Beating grafts were collected at post operation days 1, 3, 5, and 7. The erythrocyte deformability index (EDI) was measured by ektacytometry, while pathologic examinations of the hearts were performed. There were no statistically difference between group A, B, and normal group. The EDI of group C was decreased continually at 1st, 3rd, and 5th day and was statistically different from normal group at 3rd and 5th day, P <0.05, the peak of it was on the 5 th day. Group C was significantly different from group A and B, P <0.001. There were significantly correlation between EDI and pathologic results (r =0.78,P <0.001). [Conclusions] These results suggest that the erythrocyte deformation plays important roles in the course of acute rejection after heart transplantation.
